I was recently re-introduced to Pandora Radio, and I absolutely love it! Pandora allows you to customize radio stations by adding your favorite artists and songs as "seeds" which allows it to play other, similar songs which you might like. Based on what it plays, you can further customize your personal radio stations (you can have up to 100!) by thumbing up or down the individual songs which it plays. Registration is quick, free, and easy. Not sure where to start? Simply select a genre from their listings, and let the music flow!

I also discovered Last.fm recently. They actually have a better selection of music and don't seem to limit how many times you can skip songs.
